摘要: 探究染色体驱动蛋白KIF4A与胃癌细胞迁移运动的关系,从而为胃癌临床治疗提供新的治疗靶点。用G418筛选的方法构建了两种细胞株:KIF4A低表达的胃癌SGC细胞株与KIF4A过表达的胃癌SGC细胞株,Western与免疫荧光的验证后,对其分别进行细胞划痕与Transwell迁移实验检测KIF4A对细胞迁移运动能力的影响。成功构建了KIF4A低表达的SGC细胞株(SGC-shKIF4A)与KIF4A过表达的细胞株(SGC-KIF4A),在KIF4A低表达的SGC细胞株中细胞的迁移速率增加,在KIF4A过表达的细胞株中细胞的迁移速率减慢。染色体驱动蛋白KIF4A抑制胃癌细胞的迁移运动能力,为进一步研究KIF4A与胃癌转移的机制奠定基础。
